Dosage & Administration of Zybex SR 150

Smoking Cessation:

• Adult: Modified-release preparation: Start with 150 mg once daily for 6 days, then increase to 150 mg twice daily. Treatment duration: 7-12 weeks. Discontinue if abstinence is not achieved by the 7th week. Maximum: 300 mg/day.
• Elderly: 150 mg/day.
• Hepatic Impairment:
  • Mild to moderate: 150 mg once daily.
  • Severe hepatic cirrhosis: Contraindicated.

Depression:

• Adult: Initially, 100 mg twice daily, increased to 100 mg three times daily after 3 days if necessary. Further increased to 150 mg three times daily if no improvement is observed after several weeks of therapy. Maximum: 150 mg three times daily.
• Modified-release preparation: 150 mg once daily in the morning, increased to 150 mg twice daily after 3 days if necessary. Further, increase to 200 mg twice daily after several weeks if needed. Maximum: 450 mg as a single dose.
• Elderly:
  • Immediate-release tab: Initially, 37.5 mg twice daily.
  • Sustained-release tab: Initially, 100 mg daily. The dose may be increased by 37.5-100 mg every 3-4 days as tolerated. Maximum: 300 mg daily in divided doses.
• Hepatic Impairment:
  • Mild to moderate: Reduce dose or dosing frequency.
  • Severe hepatic cirrhosis (immediate-release): Maximum 75 mg once daily.
  • Severe hepatic cirrhosis (modified-release): Maximum 100 mg once daily or 150 mg every other day.

Renal Dose:

• Renal impairment: Use caution; consider dose reduction.

Interaction

Neuroleptics, lithium, TCAs, benzodiazepines, alcohol, levodopa, fluoxetine, ritonavir, and MAO inhibitors.

Contraindications

Seizure Disorder:

• Contraindicated in patients with a history of seizure disorder.
• Bupropion may induce seizures, making it unsuitable for individuals with epilepsy.

Medication Containing Bupropion:

• Contraindicated in patients using other medications containing Bupropion.
• Increased risk of seizures, and incidence is dose-dependent.

Bulimia or Anorexia Nervosa:

• Contraindicated in patients with current or prior diagnosis of bulimia or anorexia nervosa.
• Higher incidence of seizures noted in such patients treated with Bupropion Hydrochloride.

Monoamine Oxidase (MAO) Inhibitor:

• Contraindicated in concurrent use with MAO inhibitors.
• At least 14 days should elapse between discontinuation of MAO inhibitor and initiation of Bupropion Hydrochloride treatment.

Allergic Response:

• Contraindicated in patients who have shown an allergic response to Bupropion or its components.

Pregnancy & Lactation

Precautions & Warnings

• Use with caution in seizure disorders, hepatic cirrhosis, CNS tumors, and alcohol or Benzodiazepine withdrawal.
