How does Neugalin XR 82.5mg Capsule work?

Neugalin XR 82.5 is believed to work by reducing abnormal electrical activity in the brain for epilepsy, interfering with pain signals for neuropathic pain, and stopping the release of certain chemical messengers for anxiety.

Side Effects of Neugalin XR 82.5mg Capsule

Common side effects include blurred vision, difficulty paying attention, dizziness, dry mouth, edema (swelling), sleepiness, and weight gain.

• Regularly monitor for visual disturbances.
• Observe for clinical worsening, suicidality, and unusual changes in behavior, especially in those with a history of mood disorders.
• The duration of treatment depends on the condition being treated.

Indication

Pregabalin is utilized in the treatment of various conditions, including:

1. Diabetic Neuropathy: Nerve damage caused by diabetes.
2. Postherpetic Neuralgia: Persistent pain in areas previously affected by the herpes zoster virus.
3. Epilepsy: A condition characterized by abnormal brain activity, leading to seizures.
4. Fibromyalgia: A chronic condition causing widespread pain, stiffness, and other symptoms.
5. Neuropathic Pain: Pain resulting from nerve injury or damage.
6. Generalized Anxiety Disorder: Chronic worry, overthinking, and other symptoms of anxiety.

Pharmacology:

Pregabalin is a derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). Unlike direct binding to GABAA, GABAB, or benzodiazepine receptors, it exhibits a high affinity for the alpha-2-delta subunit of voltage-gated calcium channels in the central nervous system. With an oral bioavailability exceeding 90%, it is dose-independent. The drug is primarily eliminated through renal excretion in its unchanged form, featuring a half-life of approximately 6.3 hours in individuals with normal renal function.

Dosage & Administration of Neugalin XR 82.5mg Capsule

Adult Dose:

Neuropathic pain, Postherpetic Neuralgia:

• Initially: 150 mg/day
• May increase to 300 mg/day after 3-7 days.
• Maximum: 600 mg/day after a 7-day interval.
• All doses to be given in 2 or 3 divided doses.

Diabetic Peripheral Neuropathic Pain:

• Initial: 50 mg PO q8hr
• Maintenance: May increase to 100 mg PO q8hr within 1 week, as needed.
• Not to exceed 300 mg/day

Adjunct in partial seizures:

• Initially: 150 mg/day
• May increase to 300 mg/day after a week.
• Maximum: 600 mg/day.
• All doses to be given in 2 or 3 divided doses.

Fibromyalgia:

• Initially: 150 mg/day
• May increase to 300 mg/day after a week.
• Maximum: 450 mg/day, if needed.
• All doses to be given in 2 or 3 divided doses.

Child Dose: Safety and efficacy not established.

Renal Dose for Renal Impairment:

Hemodialysis:

• 25-100 mg immediately after each 4-hr hemodialysis session.

CrCl (ml/min) Dosage Recommendation:

• 30 to <60: 75 mg/day. Max: 300 mg/day. All doses to be given in 2 or 3 divided doses.
• 15 to <30: Initially, 25-50 mg/day. Max: 150 mg/day. All doses are to be given as a single dose or in 2 divided doses.
• <15: Initially, 25 mg/day. Max: 75 mg/day. All doses are to be given as a single dose.

Take Neugalin XR 82.5 as prescribed by your doctor, with or without food. It's important to take it at the same time each day. The initial dose is usually low and may be increased gradually. Missing a dose should be avoided, but if you forget, take it as soon as possible

Interaction

Lorazepam:

Report the use of Lorazepam or any other medicine that you take for sleep disorders or anxiety problems. The risk of adverse effects like dizziness, drowsiness, confusion, lack of focus, etc., is very high while taking these medicines together. Special precaution is advised if these medicines are taken by an elderly person.

Pioglitazone:

Report the use of Pregabalin to the doctor before receiving Pioglitazone or any other medicine used for controlling high blood sugar levels. These medicines could increase the risk of weight gain, fluid retention, and heart failure when taken together. You may need a dose adjustment and frequent monitoring to safely use these medicines together.

Ramipril:

Report the use of Ramipril or any other medicine belonging to the class of ACE inhibitors used for lowering blood pressure. The risk of fluid retention and associated adverse effects on the heart are significantly increased when these medicines are used together.

Oxycodone:

Report the use of any opiate pain medicines to the doctor before receiving Pregabalin. Using these medicines together can cause severe dizziness, confusion, lack of coordination especially in the elderly and people with a history of emotional disturbances, suicidal thoughts, alcohol abuse, or drug abuse. Any such effect while using them together should be reported to the doctor.

Interaction with Disease:

Angioedema:

This medicine should be administered with caution in patients who have a history of or are at risk of developing swellings of the inner layers of the skin. These swellings can cause airway blockage and prove life-threatening.
